ST056083
ST056083 is a small-molecule inhibitor of the c-di-AMP synthase DisA. The apparent KD of ST056083 binding to DisA is 21 μM. ST056083 reduces c-di-AMP production in Enterococcus faecalis ATCC 29212, and inhibits bacterial growth, biofilm formation, bacterial viability within biofilms, and the synthesis of water-insoluble extracellular polysaccharides. ST056083 is applicable to studies on bacterial c-di-AMP signaling and E. faecalis-associated infections.

ST056083 (90 or 180 μM; 48 h; 37°C; anaerobic conditions) inhibits E. faecalis biofilm formation and disrupts biofilm structure, with the effect of 180 μM being stronger than that of 90 μM[1].
ST056083 (90 or 180 μM; 48 h) reduces the average thickness of E. faecalis biofilms from 21.82 μm in the control group to 13.29 μm and 10.29 μm, respectively, and increases bacterial death within the biofilms; the dead/live ratios reach 1.73 and 2.79 at 90 and 180 μM, compared with 1.13 in the control group[1].
ST056083 inhibits c-di-AMP synthesis catalyzed by DisA, with an IC50 of 56 μM under conditions of 5 μM DisA and 50 μM ATP; when ATP is increased to 500 μM, the IC50 reaches 100 μM, and ATP at a maximum concentration of 1 mM cannot abolish the binding of ST056083 to DisA, indicating that it does not compete with ATP for DisA binding[2].
