Description

MART-1 (27-35) (human) (TFA) is the amino acid fragment spanning positions 27 to 35 of the MART-1 protein, and it represents an immunogenic epitope recognizable by HLA-A2-restricted melanoma-specific tumor-infiltrating lymphocytes (TILs). MART-1 (27-35) (human) (TFA) can be used in studies related to melanoma[1][2].

In Vitro

MART-1 (27-35) (human) (TFA) (10-7-10-5 M; 2 h at room temperature then 24 h at 4 °C) binds to purified human HLA-A2 molecules, as detected via fluorescence signal[1].
MART-1 (27-35) (human) (TFA) (10-5 M; overnight at 4 °C for initial complex formation) forms HLA-A2 complexes that dissociate almost completely within 24 h at 37 °C[1].
MART-1 (27-35) (human) (TFA) (1 μg/mL; 1 h incubation with T2 cells, 4 h cytolytic assay) is recognized by human MART-1(27-35)-specific, HLA-A2-restricted TIL clones LT8, LT11, and LT12, inducing high levels of 51Cr release from T2 target cells, with half-maximal lysis of T2 cells by clone LT12 observed at 1.1 × 10-9 M[1].
MART-1 (27-35) (human) (TFA) (10-10-10-5 g/mL; 1 h) sensitizes HLA-A*0201+ T2 cells to lysis by CLW anti-MART-1(27-35) CD8+ CTL in a dose-dependent manner, with significant lysis at 10-8 g/mL and maximum specific 51Cr release of 100% at 10-5 g/mL[2].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Cytotoxicity Assay[2]

Cell Line: HLA-A*0201+ T2 target cells, anti-MART-1(27-35) CD8+ CTL line (CLW)
Concentration: 10-10-10-5 g/mL
Incubation Time: 1 h (incubation with target cells)
Result: Induced dose-dependent specific 51Cr release from T2 cells.
Reached significant lysis at concentrations of 10-8 g/mL and above.
Achieved maximum specific 51Cr release of 100% at the highest tested concentration (10-5 g/mL).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
