Biological Activity
Cytosols are standardised to 100 μg of total protein and incubated with granzyme B from CG in different amounts adjusted to a total volume of 30 μL in 96-well flat bottom microtiter plates for 30 min at 20 °C. Following incubation with granule isolate samples, 0.4 μL N-Acetyl-Ile-Glu-Pro-Asp-p-nitroanilide (Ac-IEPD-pNA) (0.2 mM) and 70 μL of assay buffer (50 mM Hepes pH 7.4, 100 mM NaCl, 1 mM EDTA, 0.1 % NP-40, 10 % glycerol and 10 mM DTT) is added and incubated for 30 min at 37 °C. Absorption is measured at 405 nm in a microtiter plate photometry reader[1].
