ZBTB7A is a transcription factor that represses genes involved in cell proliferation and differentiation by binding to the consensus sequence 5'-[GA][CA]GACCCCCCCCC-3'. It modulates chromatin organization and recruits transcriptional regulators (e.g., HDAC1 to SMAD4-DNA complexes while blocking CREBBP/EP300 in TGF-β signaling). It interacts with factors like RELA (by similarity) and SP1 to inhibit DNA binding, acts as an AR corepressor via NCOR1/NCOR2 recruitment, and regulates fetal-to-adult globin switching via the NuRD complex. It controls glycolysis, adipogenesis (by similarity), and B/T lymphoid lineage differentiation by suppressing Notch targets (by similarity). Beyond transcription, it may participate in cNHEJ-mediated DNA repair and modulate KHDRBS1 splicing activity toward BCL2L1.
