MGB-BP-3 is an orally aective Gram-positive bactericide and DNA-binding agent. MGB-BP-3 blocks transcription initiation by binding to AT-rich bacterial DNA regions, and interferes with the functions of DNA gyrase and topoisomerase IV, but does not stabilize cleavage complexes or induce the SOS response. MGB-BP-3 induces energy depletion in *Staphylococcus aureus* and inhibits spore formation in *Clostridioides difficile*, exhibiting intrinsic resistance resilience. MGB-BP-3 can be used in the research of *Clostridioides difficile*-associated diseases .

N-(3-Oxodecanoyl)-L-homoserine lactone (3-Oxo-C10-HSL) is a bacterial signaling molecule with quorum-sensing agonistic activity and competitive antagonistic activity against short-chain AHLs. N-(3-Oxodecanoyl)-L-homoserine lactone binds to VanR to form a complex, activates the transcription of the vanI gene, and establishes an autoregulatory loop for its own synthesis. N-(3-Oxodecanoyl)-L-homoserine lactone inhibits the synthesis of violacein induced by short-chain AHLs. N-(3-Oxodecanoyl)-L-homoserine lactone serves as the sole carbon and nitrogen source for Rhodococcus erythropolis W2 and can be completely degraded. N-(3-Oxodecanoyl)-L-homoserine lactone can be used in studies related to bacterial infections .

GKL003 is a bacterial transcription inhibitor that targets the RNAP-σ interaction interface, with a Ki of 5.79 nM. GKL003 specifically binds to the RNAP β' clamp helix region at the σ A factor binding site, blocks the formation of RNAP holoenzyme, and inhibits the formation of bacterial transcription initiation complexes. GKL003 inhibits the growth of both Gram-positive and Gram-negative bacterial cells, and also exhibits activity against drug-resistant strains .

MS40 is a WDR5 PROTAC degrader with a DC50 of 42 nM in MV4;11 MLL-rearranged acute myeloid leukemia (AML) cells. MS40 induces WDR5 degradation dependent on WDR5, CRBN and the proteasome, dissociates the MLL/KMT2A complex from chromatin, reduces H3K4me2 levels, degrades IKZF1/IKZF3, the novel substrates of CRBN, inhibits the transcription of WDR5/MLL and IKZF target genes, and suppresses cancer cell growth. MS40 can be used in the research of MLL-rearranged acute myeloid leukemia, breast cancer, Burkholderia infection and cystic fibrosis-associated bacterial infections .
