β-Mannosidase is a lysosomal exo‑acting enzyme. β-Mannosidase catalyzes hydrolysis of β‑1,4‑linked mannosides, non‑reducing terminal mannoses in N‑glycoproteins, and Man‑β‑1,4‑GlcNAc linkages to release mannose. β-Mannosidase contributes to glycoprotein catabolism and supports Bifidobacteria colonization via N‑glycan depolymerization. β-Mannosidase can be used for the research of β-mannosidosis and β-mannosidase deficiency.

Biological Activity
β-Mannosidase (10 nM; 1 h) catalyzes the hydrolysis of pNP-Man, with a Km value of 1.42 mM and a kcat value of 72.2 s−1; under in vitro conditions, the activities of disease mutants W466X, R182W, G392E and S505P are significantly reduced or completely lost, while R641H, R638H and L749H retain full wild-type activity[2].
β-Mannosidase activity is undetectable in affected individuals and reduced in carriers, following autosomal recessive inheritance[3].
Purified β-Mannosidase (0.1-50 mM; 5-30 min) derived from *Bifidobacterium longum* subsp. *longum* NCC2705 efficiently and selectively cleaves the N-glycan-derived disaccharide Man-β-1,4-GlcNAc (kcat/KM = 44.3 s-1 mM-1)[4].
