In Vitro

4-Methylumbelliferyl-β-D-xylopyranoside (pH 7.0; 25 °C) is hydrolyzed by purified GSXynB2 from Geobacillus stearothermophilus T-6 with a kcat of 12.6 s−1, with the dexylosylation step of the catalytic cycle acting as the primary rate-limiting step[1].
4-Methylumbelliferyl-β-D-xylopyranoside (1.31 mM; pH 7.0; 25 °C) rapidly forms a covalent enzyme-xylose intermediate with purified GSXynB2 from Geobacillus stearothermophilus T-6, as evidenced by a presteady-state burst phase with a rate constant of 579 s−1[1].
