5-HT2A agonist-12 (Compound 4) acts as both an agonist (EC₅₀ = 0.78 μM, E_max = 68%) and a positive allosteric modulator (EC₅₀ = 0.074 μM, E_max = 53%) for the human 5-HT2A receptor. This assay is performed via a calcium mobilization experiment in Chinese hamster ovary cells expressing the receptor^[1].
