7-Coumaryl triflate
Based on 1 Customer Validation
7-Coumaryl triflate is a type 17β-HSD1 inhibitor of 17β-hydroxysteroid dehydrogenase. Its IC50 is 360 nM and Ki is 173 nM. 7-Coumaryl triflate shows selectivity for 17β-HSD2 and has no detectable affinity for ERα or ERβ. 7-Coumaryl triflate can be used in related research on hormone-dependent breast cancer.

Biological Activity
Description
In Vitro
7-Coumaryl triflate (0.6-6 μM; 10 min) potently inhibits recombinant human 17β-HSD1 overexpressed in E. coli with an IC50 of 360 nM, showing 60% and 100% inhibition at 0.6 μM and 6 μM, respectively[1].
7-Coumaryl triflate (60 μM) does not inhibit recombinant human 17β-HSD2 overexpressed in E. coli at 60 μM, confirming high selectivity for 17β-HSD1 over 17β-HSD2[1].

Cell Cytotoxicity Assay
Cytotoxicity assays are usually based on the assessment of cell membrane damage, which can also be indirectly detected by measuring cell viability. Detection methods include MTT assay, CKK-8 assay, LDH assay and ATP assay, etc.

Research Protocol for Endocrine Diseases
Endocrine diseases often arise from disrupted hormone production, hormone signaling, or target-tissue responsiveness; for diabetes-focused endocrine disease models, insulin signaling regulates glucose uptake, hepatic glucose output, lipid metabolism, and β-cell compensation. Type 2 diabetes develops through interacting defects in insulin resistance, β-cell dysfunction, adipose inflammation, hepatic glucose overproduction, altered incretin signaling, and ectopic lipid metabolism. A major unresolved question is whether endocrine dysfunction is driven primarily by target-tissue insulin resistance, intrinsic β-cell failure, immune/inflammatory stress, or combined multi-organ failure that differs by disease stage.

Breast Cancer Modeling
Breast cancer is a heterogeneous cancer, and it has been distinguished into four subtypes: luminal A, luminal B, HER2-positive and basal-like. Molecular mutations, epigenetic alterations, hormone exposure and immune microenvironment are related to the progression of breast cancer.
