Function
CBLB/RNF56 Protein serves as an E3 ubiquitin-protein ligase, mediating the transfer of ubiquitin from specific E2 ubiquitin-conjugating enzymes to substrates, ultimately promoting their degradation by the proteasome. It plays a pivotal role in negatively regulating signal transduction pathways initiated by TCR, BCR, and FCER1. In naive T-cells, CBLB inhibits VAV1 activation upon TCR engagement and mandates CD28 costimulation for proliferation and IL-2 production. Furthermore, it promotes ubiquitination of PIK3R1/p85, hindering its recruitment to the TCR and subsequent activation. In activated T-cells, CBLB hampers PLCG1 activation and calcium mobilization upon restimulation, contributing to the induction of anergy. In B-cells, it ubiquitinates SYK, facilitating its proteasomal degradation. Additionally, CBLB may play a role in SRC ubiquitination and could be involved in EGFR ubiquitination and internalization. It is suggested to work in conjunction with the E2 ubiquitin-protein ligase UB2D3 and, when associated with CBL, is essential for the proper feedback inhibition of ciliary PDGFRA signaling through ubiquitination and internalization of PDGFRA.

Tissue specificity: Expressed in placenta, heart, lung, kidney, spleen, ovary and testis, as well as fetal brain and liver and hematopoietic cell lines, but not in adult brain, liver, pancreas, salivary gland, or skeletal muscle. Present in lymphocytes (at protein level).

CBLB has three isomers: Long: 982 amino acids, molecular weight 109,450 Da (predicted); Truncated 1: 810 amino acids, molecular weight 90,754 Da (predicted); Truncated 2: 770 amino acids, molecular weight 86,582 Da (predicted).
After TCR or BCR activation, and under various cellular stimuli, tyrosine and serine residues are phosphorylated. EGF-mediated cell activation or CD28 co-stimulation of T cells leads to autoubiquitination, thereby promoting proteasome degradation. -

Interacts with SH3 domain-containing proteins LCK, CRK and SORBS1. Interacts with LCP2 and ZAP70. Interacts with CBL. Interacts with SH3 domain-containing proteins VAV1, FYN, FGR, PLCG1, GRB2, CRKL, PIK3R1 and SH3KBP1/CIN85. Identified in heterotrimeric complexes with SH3KBP1/CIN85, CD2AP and ARHGEF7, where one CBLB peptide binds two copies of the other protein. Interacts with poly-ubiquitinated proteins. Dimerization is required for the binding of poly-ubiquitin, but not for the binding of mono-ubiquitin. Interacts with EGFR (phosphorylated). Interacts with IFT20.
