Function
Signaling Lymphocytic Activation Molecule Family Member 2 (SLAMF2, CD48, BLAST-1) is a glycosylphosphatidylinositol (GPI)-anchored cell-surface glycoprotein that regulates immune cell function and activation by interacting with cell-surface receptors (such as CD244/2B4 or CD2) via its N-terminal immunoglobulin domain. CD48 participates in T-cell signal transduction by binding to CD2 and effectively recruiting Src-family protein kinases LCK and LAT to the TCR/CD3 complex. CD48 induces phosphorylation of the immunoreceptor tyrosine-based switch motif (ITSM) within the cytoplasmic region of CD244, thereby initiating a signaling cascade that leads to the formation of the immunological synapse and triggers the directed release of cytotoxic granules-containing perforin and granzymes-from T lymphocytes and NK cells.
