  • Western blot analysis of extracts from SH-SY5Y(lane 2(20ug) , SH-SY5Y(lane 3(40ug) and HEK293T(lane 4(20ug) , HEK293T(lane 5(40ug) using Periostin Antibody (HY-P81535) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background
Function：Induces cell attachment and spreading and plays a role in cell adhesion (PubMed:12235007). Enhances incorporation of BMP1 in the fibronectin matrix of connective tissues, and subsequent proteolytic activation of lysyl oxidase LOX (By similarity)
Subcellular Localization：Golgi apparatus; Secreted; Secreted, extracellular space, extracellular matrix
Expression：
Tissue_specificity:It is widely expressed, with the highest expression levels in the aorta, stomach, lower digestive tract, placenta, uterus, thyroid tissue, and breast. It is also expressed in the kidneys (PubMed: 21763681) and lungs (PubMed: 22079858) . Its expression is upregulated in ovarian epithelial tumors. It is not expressed in normal ovaries. It is also highly expressed around the tumor in lung cancer tissue, but not within the tumor itself. It is overexpressed in breast cancer.
Isoforms & Post-Translational Modification：Q15063 has 10 isomers: Q15063-1: 93314 Da (predicted); Q15063-2: 87021 Da (predicted); Q15063-3: 87254 Da (predicted); Q15063-4: 83850 Da (predicted); Q15063-5: 90424 Da (predicted); Q15063-6: 83527 Da (predicted); Q15063-7: 80357 Da (predicted); Q15063-8: 90143 Da (predicted); Q15063-9: 90010 Da (predicted); Q15063-10: 86840 Da (predicted).
Gamma-carboxylation is controversial. Gamma-carboxyglutamated; gamma-carboxyglutamate residues are formed by vitamin K dependent carboxylation; this may be required for calcium binding (PubMed:18450759). According to a more recent report, does not contain vitamin K-dependent gamma-carboxyglutamate residues (PubMed:26273833)
