FGL peptide is a fibroblast growth factor receptor (FGFR) modulator and blood-brain barrier-penetrant. FGL peptide activates NCAM-FGFR and FGFR1 signaling pathways. FGL peptide alters expression of apoptosis, signal transduction and metabolism regulator genes in traumatic brain injury contexts. FGL peptide can be used for the research of traumatic brain injury.

In Vivo
FGL peptide (2.6-26.0 µg; i.n.; on PND1, 2, and 3) accelerates maturation of vestibular-dependent sensorimotor coordination skills in newborn Wistar rats, specifically reducing surface righting reflex latency and increasing successful negative geotaxis responses on PND4-6, without affecting general motor activity, muscle strength, or body weight[2].

Result:Altered the expression of 82 genes via interaction with TBI in injured animals. Upregulated a gene cluster including Fas apoptotic inhibitory molecule, regulator of G-protein signaling 14, and solute carrier family 7 member 1 at 4 days post-injury. Modulated expression of genes involved in metabolism, transcription, and cytoskeleton maintenance, with some genes showing upregulation at 6 hours and 4 days but downregulation at 1 day post-injury. Upregulated genes encoding neurofilament, adenylate cyclase activating polypeptide 1, and RNA binding protein p45AUF1 in non-injured animals. Downregulated genes encoding RNA helicase II, mitochondrial ribosomal protein 9, and metaxin 2 in non-injured animals.

TUNEL staining for apoptotic DNA fragmentation
TUNEL staining detects DNA strand breaks by using terminal deoxynucleotidyl transferase to add labeled nucleotides to exposed 3′-OH DNA termini, generating either microscopic staining in fixed cells or tissue sections, or fluorescence/cytometric signal in cell suspensions. TUNEL positivity reflects DNA fragmentation but should not be interpreted alone as definitive apoptosis, because TUNEL can also label necrotic, autolytic, mechanically damaged, or DNA-repair-associated DNA breaks.

Annexin V plus membrane-impermeant dye apoptosis staining
Annexin V-based apoptosis assays rely on the detection of phosphatidylserine (PS) externalization from the inner leaflet of the plasma membrane to the outer leaflet, an early biochemical hallmark of apoptosis. Fluorescently labeled Annexin V binds PS in a calcium-dependent manner, enabling identification of early apoptotic cells by flow cytometry or fluorescence microscopy. When combined with a membrane-impermeant DNA-binding dye (e. g. , propidium iodide), this approach allows discrimination between viable (Annexin V−/dye−), early apoptotic (Annexin V+/dye−), and late apoptotic or necrotic (Annexin V+/dye+) cell populations by assessing membrane integrity and PS exposure.
