TAF1
TAF1 is the largest subunit of the general transcription factor TFIID and one of the most multifunctional proteins in this atypical-kinase collection. It harbors Ser/Thr kinase activity toward histone H1 and toward TAF1 itself, intrinsic histone acetyltransferase activity, ubiquitin-activating and -conjugating activities, and two bromodomains that read acetylated histones. Through these modules TAF1 integrates promoter recognition with chromatin modification during RNA polymerase II initiation. An SVA retrotransposon insertion and point mutations in TAF1 cause X-linked dystonia-parkinsonism (DYT3). Recombinant TAF1 and its domains support transcription reconstitution, chromatin-enzyme assays and neurological disease modeling.
