Disuprazole
Disuprazole (U-69167) is an orally active benzimidazole proton pump inhibitor that inhibits gastric H+/K+-ATPase with an IC50 of 2 × 10-5 M. Disuprazole reduces gastric acid secretion by inhibiting gastric proton pump activity. Disuprazole can be used in research related to gastric acid secretion and peptic ulcer.

In Vitro
Disuprazole (10 ns) binds to CYP2C9 in an extended conformation 10~15 Å above the heme, and its binding site is stabilized by a water-mediated hydrogen-bonding network and strong hydrophobic interactions[2].
Disuprazole inhibits hog gastric (H+-K+)-ATPase activity with an IC50 of 2 × 10−5 M[1].

In Vivo
Disuprazole (s.c. or oral administration; single dose) inhibits gastric acid secretion in Carbachol (HY-B1208)-stimulated pylorus-ligated Shay rats, with ED50 values of < 1.0 mg/kg and 2.0-5.0 mg/kg, respectively[1].
