SA 47 is a selective fatty acid amide hydrolase (FAAH) inhibitor. SA 47 inhibits FAAH enzyme activity and disrupts the NLRP3-FAAH interaction, promoting K48 ubiquitination and selective autophagic degradation of NLRP3. SA 47 reduces NLRP3 protein levels in macrophages. SA 47 can be used for research on inflammatory diseases associated with NLRP3 protein mutations.

SA 47 (40 μM) induces NLRP3 protein degradation in BMDMs, reducing NLRP3 protein levels by more than 30%[1].
SA 47 (10 μM; 30 min) does not inhibit any of the seven cloned rat carboxylesterases in HEK293T cells (X51974/TGH/ES10, NM_031565/ES3, NM_133586/ES2, NM_144743, NM_017004, X81825/ES4, and NM_001024365)[2].
SA 47 (10 μM; 30 min) does not significantly inhibit carboxylesterase activity in rat liver microsomes or cytosol[2].
SA 47 shows excellent selectivity for FAAH in both mouse and human tissues, with no significant off-target effects at 10 μM[2].

Autophagy
Autophagy is a process in which eukaryotic cells use lysosomes to degrade their own cytoplasmic proteins and damaged organelles under the regulation of autophagy related gene (Atg). Microtubule-associated proteins light chain 3 (LC3) is recognized as autophagy marker, which transfers from cytoplasmic LC3 (LC3-I) to membrane type (LC3-II). LC3-II/I ratio could be detected by Western Blot and fluorescence microscopy.
