Disuprazole (10 ns) binds to CYP2C9 in an extended conformation 10~15 Å above the heme, and its binding site is stabilized by a water-mediated hydrogen-bonding network and strong hydrophobic interactions^[2].
Disuprazole inhibits hog gastric (H⁺-K⁺)-ATPase activity with an IC₅₀ of 2 × 10⁻⁵ M^[1].

In Vivo

Disuprazole (U-69167) (compound 26) (s.c. or p.o.; single administration; 3 h) inhibits gastric H⁺/K⁺-ATPase activity in male Sprague-Dawley rats with ED₅₀ values of 1.0 mg/kg and 15 mg/kg, respectively^[1].
Disuprazole (s.c. or oral administration; single dose) inhibits gastric acid secretion in Carbachol (HY-B1208)-stimulated pylorus-ligated Shay rats, with ED₅₀ values of < 1.0 mg/kg and 2.0-5.0 mg/kg, respectively^[1].
