MOA-stilbene

MOA-stilbene is an inhibitor targeting mitochondrial complex III, with fungicidal and insecticidal activities. MOA-stilbene binds to the Qo center of cytochrome b (i.e., the ubiquinol oxidation site, center o), blocking electron transport and inhibiting mitochondrial cellular respiration, thereby reducing the production of reactive oxygen species. MOA-stilbene binds to the Qi center of the chloroplast b6f-complex, inhibiting the reoxidation of reduced bH and bL hemes, the reverse electron transfer from cytochrome c + c1 and ISP to bH and bL, and the electron transfer from bH to bL.

Oxidative Stress

In COS-7 cells and SMP, MOA-stilbene (1 μM; 1 h) produces hydrogen peroxide levels similar to control values under basal conditions; however, in glucose-supplemented COS-7 cells, MOA-stilbene substantially decreases the rate and extent of hydrogen peroxide production, especially in the presence of C6-ceramide^[1].

Bacterial Infection

MOA-stilbene is a potent inhibitor of mitochondrial respiration in bovine heart mitochondria, exhibiting an I₅₀ of 0.04 mM^[2].
In bovine SMP, MOA-stilbene (5 mM) in combination with antimycin causes less inhibition of succinate-induced b heme reduction than the combination of antimycin and myxothiazol^[3].

MOA-stilbene inhibits complex III from yeast, Botrytis, fly, rat, and corn with pI₅₀ values of 7.3, 7.4, 7.8, 7.6, and 6.3, respectively^[2].
