Neurofascin
Neurofascin, encoded by NFASC, is an L1 family adhesion molecule that exists as two principal isoforms generated by alternative splicing: the glial NF155 and the neuronal NF186. NF155 is expressed by myelinating glia and, together with contactin-1 and Caspr, forms the paranodal axoglial junction that seals myelin loops against the axon, while NF186 clusters at nodes of Ranvier where it helps organize voltage-gated sodium channels with ankyrin-G. Autoantibodies against NF155 define a severe subtype of chronic inflammatory demyelinating polyradiculoneuropathy that responds poorly to standard therapy but improves with B cell-targeted treatment, making neurofascin a clinically important neuroimmunology antigen.
