Background

The ADAM17/TACE protein functions as a pivotal protease involved in the cleavage of various membrane-bound precursor proteins, contributing to the regulation of diverse biological processes. Notably, ADAM17/TACE cleaves the membrane-bound precursor of TNF-alpha, releasing its mature soluble form, and is responsible for the proteolytic release of soluble JAM3 from endothelial cell surfaces. Additionally, it plays a critical role in the proteolytic release of several cell-surface proteins, including p75 TNF-receptor, interleukin 1 receptor type II, p55 TNF-receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor, MUC1, and the amyloid precursor protein. ADAM17/TACE serves as an activator of the Notch pathway by mediating the cleavage of Notch, generating the membrane-associated intermediate fragment called Notch extracellular truncation (NEXT). Furthermore, it participates in the proteolytic processing of ACE2 and plays a role in hemostasis through the shedding of GP1BA, the platelet glycoprotein Ib alpha chain. ADAM17/TACE also mediates the proteolytic cleavage of LAG3 and IL6R, leading to the release of their secreted forms, and facilitates the shedding of FCGR3A upon NK cell stimulation, promoting increased NK cell motility and detachment from opsonized target cells.
