Biological Activity
Background
CD86 is a transmembrane region, and a longer cytoplasmic domain than CD80. CD86 is constitutively expressed on interdigitating dendritic cells (DCs), Langerhans cells, peripheral blood DCs, memory B cells and germinal center B cells, and macrophages. CD86 is rapidly upregulated on B cells following activation by cross-linking of the Ig receptor or the addition of a variety of cytokines[1].
Verified Bioactivity
1.Immobilized Human B7-2 at 5 μg/mL (100 μL/well) can bind Biotinylated Human CTLA-4 (HY-P78890). The ED50 for this effect is 15.19 ng/mL.
2.Immobilized Biotinylated Mouse CD28 (HY-P704990) at 1 μg/mL (100 μL/well) on Streptavidin precoated (0.5 μg/well) plate can bind Human B7-2. The ED50 for this effect is 9.828 ng/mL.
3.Human B7-2 Protein Immobilized on Protein A Chip can bind CD28 (HY-P70486) Protein with an affinity constant of 14.6 μM as determined in SPR assay.
4.Human B7-2 Protein Immobilized on Protein A Chip can bind Human CTLA-4 (HY-P70684) with an affinity constant of 3.88 nM as determined in SPR assay.
5.Flow Cytometry assay shows that Human B7-2 can bind to CD28 expressed on Jurkat. The concentration of B7-2 used is 2 μg/mL.
