Background

The C5AR1 Protein-VLP serves as a receptor for the chemotactic and inflammatory peptide anaphylatoxin C5a. The ligand engages with at least two distinct sites on the receptor: a high-affinity site located on the extracellular N-terminus and a second site within the transmembrane region, initiating downstream signaling events. Upon receptor activation, a spectrum of cellular responses is triggered, including chemotaxis, granule enzyme release, intracellular calcium release, and superoxide anion production. The receptor can exist as a homodimer and may form higher-order oligomers. When phosphorylated, C5AR1 interacts with ARRB1 and ARRB2, facilitating the internalization of C5aR. Additionally, the N-terminal domain of C5AR1 engages with S. aureus chemotaxis inhibitory protein (CHIPS), potentially inhibiting the immune response by blocking the receptor. This intricate molecular interplay underscores the multifaceted role of C5AR1 in orchestrating immune and inflammatory processes.
