Assay Procedure
Materials
Assay buffer: 12.5 mM Tris, 75 mM NaCl, pH 7.5
Test protein: Carbonic Anhydrase 9 Protein, Human (HEK293, His) (HY-P70064)
Substrate: 4-Nitrophenyl Acetate
Procedure
1. Dilute Carbonic Anhydrase 9 to 20 ng/μL in the assay buffer.
2. Dilute the substrate to 2 mM in the assay buffer.
3. Load 50 μL of 20 ng/μL Carbonic Anhydrase 9 into the plate, and start the reaction by adding 50 μL of 2 mM substrate to the well.
4. Include a substrate blank containing 50 μL of assay buffer and 50 μL of 2 mM substrate.
5. Read the absorbance at 400 nm in kinetic mode (bottom reading) for 5 minutes.
6. Calculate the specific activity:
