Biological Activity

Description

CARD11 Protein, Human (His, Strep) is the recombinant human-derived CARD11, expressed by E. coli , with Strep, His labeled tag.

Background

CARD11 is an adapter protein that plays a key role in adaptive immune response by transducing the activation of NF-kappa-B downstream of T-cell receptor (TCR) and B-cell receptor (BCR) engagement (by similar). It transduces signals downstream of TCR or BCR activation via the formation of a multiprotein complex with BCL10 and MALT1, inducing NF-kappa-B and MAP kinase p38 (MAPK11, MAPK12, MAPK13, and/or MAPK14) pathways (by similar). Upon TCR or BCR triggering, CARD11 homooligomerizes to form a helical template recruiting BCL10 via CARD-CARD interaction, promoting BCL10 polymerization and subsequent MALT1 recruitment, leading to I-kappa-B kinase (IKK) phosphorylation, degradation, and NF-kappa-B nuclear translocation. Its binding to DPP4 induces T-cell proliferation and NF-kappa-B activation in a TCR/CD3-dependent manner (by similar). CARD11 promotes linear ubiquitination of BCL10 by targeting it to RNF31/HOIP and stimulates BCL10 phosphorylation (by similar). It also activates the TORC1 signaling pathway.
