Background
LDLR protein functions by binding to LDL, the primary lipoprotein responsible for carrying cholesterol in the bloodstream, and facilitating its uptake into cells through endocytosis. For internalization to occur, the receptor-ligand complexes must cluster within clathrin-coated pits. It engages in interactions with various proteins, including DAB2 (via PID domain) through the NPXY motif, although tyrosine phosphorylation of the NPXY motif can hinder this interaction. LDLR also interacts with LDLRAP1 (via PID domain) via the NPXY motif, as well as with ARRB1 and SNX17.
