Biological Activity
OXCT1 Protein, Human (His) is the recombinant human-derived OXCT1, expressed by E. coli , with N-6*His labeled tag. ,
OXCT1 is a key enzyme in ketone body catabolism, catalyzing the first and rate-limiting step of ketone body utilization in extrahepatic tissues. It transfers coenzyme A (CoA) from a donor thiolester (succinyl-CoA) to an acceptor carboxylate (acetoacetate), producing acetoacetyl-CoA. Acetoacetyl-CoA is further metabolized by acetoacetyl-CoA thiolase into two acetyl-CoA molecules, which enter the citric acid cycle for energy production. The enzyme forms a dimer where both subunits can form enzyme-CoA thiolester intermediates, but only one subunit is capable of transferring the CoA moiety to the acceptor carboxylate (3-oxo acid) to generate a new acyl-CoA. The formation of the enzyme-CoA intermediate occurs via an unstable anhydride species between the carboxylate groups of the enzyme and substrate (By similarity).
