Background
PAK2 is a serine/threonine protein kinase involved in diverse signaling pathways regulating cytoskeleton dynamics, cell motility, cell cycle progression, apoptosis, and proliferation. It acts as a downstream effector of CDC42/RAC1 GTPases, requiring binding to active CDC42/RAC1 for conformational change and autophosphorylation. Full-length PAK2 promotes cell survival/growth, phosphorylates MAPK4/MAPK6 to activate MAPKAPK5 (regulating F-actin polymerization and migration), and mediates EGF-induced proliferation via JUN phosphorylation. Other substrates include histone H4 (nucleosome assembly), BAD, S6 ribosomal protein, and MBP. PAK2 inhibits CASP7 activity through phosphorylation and performs kinase-independent roles (e.g., mitotic spindle orientation) with ARHGEF7/GIT1. Under apoptotic stimuli (e.g., DNA damage), caspase-cleaved PAK2 generates PAK-2p34, which translocates to the nucleus to drive apoptosis via JNK signaling; caspase-activated PAK2 also phosphorylates MKNK1 to suppress translation.
