Background
TGF alpha, a mitogenic polypeptide, exhibits the ability to bind to the EGF receptor (EGFR) and synergistically collaborate with TGF beta to facilitate anchorage-independent cell proliferation in soft agar. It interacts with the PDZ domains of SDCBP and SNTA1, with the former being crucial for its localization to the cell surface. In its immature form, present in the endoplasmic reticulum and characterized by a prosegment and partial N-glycosylation, TGF alpha interacts with CNIH. Within the Golgi apparatus, it may form a complex with CNIH and GORASP2. Additionally, TGF alpha interacts with NKD2 through its cytoplasmic C-terminal domain and with MAGI3.
Verified Bioactivity
Measured in a cell proliferation assay using Balb/3T3 mouse embryonic fibroblast cells. The ED50 for this effect is <0.4 ng/mL.
