RNase L Recruiter-PEG3-acid (4 M HCl in 1,4-dioxane; 1 h; room temperature) is obtained from RNase L Recruiter-PEG3-COOtBu via acidic deprotection; LC-MS shows complete deprotection, and the resulting crude white solid is used in the next reaction without further purification^[1].
RNase L Recruiter-PEG3-acid (1.0 equiv; HATU 1.2 equiv; DIPEA 4.0 equiv; 5 min preactivation; room temperature; 2 h) undergoes amide coupling with 2-(3-(but-3-yn-1-yl)-3H-diazirin-3-yl) ethan-1-amine. After complete reaction, the downstream product RNase L Recruiter Diaz is obtained as a white solid via RP-HPLC purification in 65% yield^[1].
