DMF-DMA (N-Dimethoxymethyl-N,N-dimethylamine) is a reagent with both condensation and alkylation functions. DMF-DMA can undergo condensation reactions with active methylene groups or amino groups to generate enamine and imino derivatives, and can also act as an alkylating agent to methylate nitrogen and oxygen atoms in heterocyclic structures. MitoPerOx (solution) is a mitochondrial-targeted, lipid peroxidation-indicating fluorescent probe with BODIPY581/591 fluorophores. The triphenylphosphine cation (TPP+) of MitoPerOx can be selectively enriched in mitochondria (depending on membrane potential) and can be used to detect lipid peroxidation in the inner mitochondrial membrane. Under the action of lipid peroxides, the BODIPY581/591 fluorophores of MitoPerOx shift their emission wavelength from 590 nm (reduced state) to 520 nm (oxidized state) , and ratiometric detection can be performed at an excitation wavelength of 488 nm. Tributyl citrate is a low-toxicity and orally active citrate ester with no genotoxicity or skin sensitizing activity. Tributyl citrate also acts as a plasticizer, solvent, FDA-approved indirect food additive, and topical anesthetic, among other uses. Tributyl citrate induces a needle-prick insensitivity response that lasts for more than 2 hours, and a 5% suspension of it temporarily eliminates the corneal reflex in rabbits. SL-052 is a hypocrellin-based photosensitizer that has recently shown promising results in clinical and preclinical testing for cancer photodynamic therapy (PDT). SL-052 is encapsulated in biodegradable polylactic-co-glycolic acid (PLGA) polymer nanoparticles optimized using single emulsion solvent evaporation technology. The SL-052-PLGA nanoparticles were more effective in PDT treatment of subcutaneous SCCVII squamous cell carcinoma compared to polyvinylpyrrolidone (PVP)-based and standard liposomal SL-052 formulations. A longer time interval between drug injection and tumor illumination can improve tumor cure rates, and SL-052-PLGA nanoparticles showed the best therapeutic effect among all SL-052 formulations.

Prodan is an environment-sensitive fluorescent probe used to investigate the polarity, fluidity, and structure of lipid membranes. Prodan exhibits environment-dependent excitation/emission wavelengths, with its wavelength range covering Ex = 360-410 nm and Em = 430-530 nm when in solvents, bound to DNA, located in lipid bilayers, or associated with proteins. Prodan can interact with the major groove of DNA, regions of lipid bilayers, and hydrophobic pockets of proteins. During phase transitions, Prodan localizes to different regions of the bilayer: the emission minimum is 430-440 nm in the gel phase, while it shifts to 480-500 nm in the liquid-crystalline phase or interdigitated gel phase. Mito-polarity is a mitochondria-targeted polarity probe. Mito-polarity can be used to detect mitochondrial polarity. The detection mechanism of Mito-polarity is based on an intramolecular charge transfer (ICT) system designed with a donor-π-bridge-acceptor (D-π-A) structure. Among them, the 467 nm green emission generated by the Coumarin (HY-N0709) structure is extremely sensitive to polarity changes, while the 642 nm red emission produced by the extended π-conjugation and ICT effect between the Coumarin and benzothiazine structures shows only a weak response, thus forming a ratiometric fluorescence response that is linearly correlated with solvent polarity. When Mito-polarity is excited at 405 nm in methanol, its excitation/emission wavelengths are Ex/Em = 405/467 nm and Ex/Em = 405/642 nm, with maximum absorption wavelengths of 426 nm and 561 nm .

Nile-TPA is a fluorescent probe for polarity detection, which can be used for lipid droplet targeting, cell polarity analysis, in vivo polarity imaging, and blood polarity monitoring in circulating tumor mouse models. The detection mechanism of Nile-TPA relies on the dual intramolecular charge transfer (ICT) effect generated by the electron-donating triphenylamine and Nile Red moieties and the electron-withdrawing carbonyl group; this probe emits strong red fluorescence in a low-polarity environment, and as polarity increases, the fluorescence intensity decreases and a red shift occurs due to changes in the excited-state dipole moment.
