TPA-IM
TPA-IM is an Aβ40-binding inhibitor with a Ka of 12.7 nM. TPA-IM inhibits amyloid-β fibrillation and disrupts preformed fibrils. TPA-IM inhibits intracellular ROS production, restores mitochondrial membrane potential, attenuates amyloid-β-induced apoptosis (apoptosis), and rescues amyloid-β-induced loss of neuronal viability. TPA-IM self-assembles into spherical nanoaggregates and co-assembles with amyloid-β, redirecting aggregation toward non-fibrillar structures. TPA-IM can be used for research on Alzheimer's disease.

Biological Activity
Description
In Vitro
TPA-IM (30 μM) self-assembles into spherical nanoaggregates of approximately 100-150 nm and co-assembles with Aβ40 into non-fibrillar hybrid structures[1].
TPA-IM (0-100 μM; 48 h) shows no cytotoxicity in SH-SY5Y and IMR-32 cells up to 100 μM and rescues Aβ40-induced loss of viability in SH-SY5Y cells[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Line:SH-SY5Y and IMR-32 human neuroblastoma cells
-
Concentration:0, 10, 20, 30, 40, 50, 75, 100 μM (TPA-IM); 10-100 μM (MTT interference study)
-
Incubation Time:48 h (TPA-IM); 2 h (MTT); 15 min (DMSO)
-
Result:Exhibited negligible intrinsic cytotoxicity in SH-SY5Y cells over 1-100 μM, maintaining high cellular viability even at elevated doses.
In IMR-32 cells, TPA-IM maintained comparatively higher cell viability.
Aβ40 fibrils dose-dependently reduced neuronal viability; co-incubation with TPA-IM significantly rescued cell viability relative to the Aβ40 group.
