trans-Zeatinriboside is a zeatin-type cytokinin and plant growth regulator. trans-Zeatinriboside is produced by the terrestrial filamentous cyanobacterium Nostoc sp. HK-01 and Agrobacterium tumefaciens (including Ti plasmid-dependent and Ti plasmid-independent strains). trans-Zeatinriboside is detectable as a cytokinin component in growing crown galls, and accumulates in large amounts in CHRK1-silenced transgenic tobacco accompanied by pleiotropic developmental abnormalities. trans-Zeatinriboside can be used in studies related to crown galls.

Compared with wild-type seedlings, the endogenous trans-Zeatinriboside level increases 5.9-fold in 10-day-old CHRK1-silenced transgenic Nicotiana tabacum cv. Xanthi seedlings, and 3.0-fold in CHRK1-silenced transgenic Nicotiana tabacum cv. Xanthi clustered calli; meanwhile, the total cytokinin level in transgenic tissues increases 3-fold[2].
trans-Zeatinriboside exhibits extremely low cross-reactivity (only 1.8%) with the anti-isopentenyladenosine antiserum used in radioimmunoassay[3].
trans-Zeatinriboside is produced at detectable low levels in stationary-phase liquid cultures of the virulent Agrobacterium tumefaciens strain B6 and the avirulent strain B6-37[3].
trans-Zeatinriboside (at 48 h, 360 h, 500 h, 7 days, and 21 days after inoculation) significantly increases the level of trans-zeatin riboside-equivalent cytokinins in established crown galls across multiple angiosperm species, including Euphorbia lathyris, Helianthus annuus, Sinapis alba, and Catharanthus roseus, with the maximum increase reaching up to 190-fold (Euphorbia lathyris); however, this elevation is not observed in some tumor-forming species after infection with Agrobacterium tumefaciens B6[3].
The content of trans-Zeatinriboside varies drastically among bacteria-free crown gall callus cultures grown on hormone-free medium, ranging from undetectable (Nicotiana tabacum) to 429 nmol/g DW (Tavaresia glandiflora)[3].

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.08 mg/mL (5.92 mM); Clear solution
This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
