TRITC-dextran MW 70000 (Tetramethyl rhodamine isothiocyanate glucan, MW 70000) is a fluorescent dye with a molecular weight of 70 kD. Paracellular Permeability Assay[1]
1. Add TRITC-dextran MW 70000 (20 mg/mL, 100 μL) to the basal medium in the upper chamber of a transwell (pore size 0.4 μm).
2. Collect the medium from the lower transwell chamber at 0, 30, 60, and 120 minutes.
3. Measure the fluorescence signal (Ex=544 nm, Em=570 nm).
4. Calculate the concentration of TRITC-dextran MW 70000 based on the fluorescence intensity.
5. Calculate the permeability rate.
