Description

2-Deoxyribose 5-phosphate disodium is a substrate of 2-deoxyribose-5-phosphate aldolase (DERA). DERA belongs to class I aldolases, catalyzes reversible aldol reactions, and does not require any cofactors[1].

Guidelines (The following is our recommended protocol, which serves only as a guideline and should be modified according to your specific needs)[1].
DERA Activity Assay.
1. Prepare 2-Deoxyribose 5-phosphate disodium working solution: Prepare 1 mM 2-Deoxyribose 5-phosphate disodium in 20 mM Na2HPO4-citrate buffer (pH 7.0).
2. Experimental Procedures:
2.1 The reaction system contains 20 mM Na2HPO4-citrate buffer (pH 7.0), 1 mM substrate (2-Deoxyribose 5-phosphate disodium or 2-deoxyribo), 0.5 mM NADH, 6 U triosephosphate isomerase (EC 5.3.1.1, TPI), 2 U glycerophosphate dehydrogenase (EC 1.1.1.8, GDP) and 10 μL diluted DERA, with a total volume of 200 μL.
2.2 The reaction proceeds at 35 °C for 30 min.
2.3 A universal microplate spectrophotometer is used to measure the decrease in absorbance at 340 nm caused by NADH consumption. The consumption of one NADH molecule corresponds to the decomposition of one DRP molecule. One enzyme unit (U) is defined as the amount of enzyme required to catalyze the cleavage of 1 μmol of substrate per minute under the assay conditions.
2.4 Protein concentration is quantified using a BCA Protein Assay Kit according to the method reported in the literature.
