Description

Species Reactivity

Human

In Vitro

23ME-01473 (threefold serial dilutions; 37°C) binds to purified hULBP6, hULBP2 and hULBP5 proteins with high affinity, among which it shows the strongest affinity for hULBP6 (KD = 0.053 nmol/L), and does not bind to ULBP1, ULBP3 or ULBP4[1].
23ME-01473 (three-fold serial dilution; room temperature) potently blocks the binding of purified ULBP5 and ULBP6-02 to purified NKG2D, with a stronger inhibitory effect on ULBP6-02 (IC50 = 0.04 nmol/L) than on ULBP5 (IC50 = 0.31 nmol/L)[1].
23ME-01473 (threefold serial dilutions; 37°C) binds to purified activated hFcγ receptors with high affinity, particularly FcγRIIIa-V176 (KD = 6.74 nmol/L) and FcγRIIIa-F176 (KD = 25.50 nmol/L), while it shows low affinity for the inhibitory FcγRIIb[1].
23ME-01473 (0.05-200 nmol/L) significantly enhances IFNγ production by hPBMCs pre-stimulated with IL-2/IL-15 and co-cultured with COV644 cells and soluble ULBP6-02, and its activity is superior to that of the Fc-WT anti-ULBP6/ULBP2/ULBP5 variant[1].
23ME-01473 (100 nmol/L; 5 days) potently enhances the cytotoxicity of IL-2/IL-15-prestimulated hPBMC against COV644-GFP ovarian cancer cells in the presence of soluble ULBP6-02, and its activity is superior to that of effector function-impaired antibodies and isotype control antibodies[1].
23ME-01473 (0.0001-10 nmol/L; per assay protocol) potently induces antibody-dependent cell-mediated cytotoxicity (ADCC) against COV644 ovarian cancer cells via activating FcγRIIIa, and its activity is significantly higher than that of the Fc-WT anti-ULBP6/2/5 variant[1].

In Vivo

23ME-01473 (10 mg/kg, twice weekly for 6 weeks) induces tumor growth inhibition in NSCLC PDX models implanted with hNK cells[1].

Animal Model: hIL-15 NOG mice (female, Taconic Inc.)[1]
Dosage: 10 mg/kg
Administration: twice weekly; 6 weeks
Result: Achieved 48% tumor growth inhibition compared with isotype control.
Showed slower tumor growth kinetics, particularly near the end of the dosing regimen, compared with control group.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
