3-(N-Maleimidopropionyl)biocytin is a cysteine-specific labeling reagent and non-membrane-permeable probe. 3-(N-Maleimidopropionyl)biocytin covalently modifies the exposed cysteine residues of yeast Tim44, conjugating the biotin moiety to the polypeptide chain. 3-(N-Maleimidopropionyl)biocytin determines the topology of membrane proteins by modifying the exposed cysteine residues on the outer side of the inner mitochondrial membrane[1].

In Vitro

3-(N-Maleimidopropionyl)biocytin (0.5 mM; 30 min at 4 °C) labels the single cysteine residue (Cys-369) of yeast Tim44 in intact mitochondria, demonstrating that the C-terminal domain of yeast Tim44 is exposed to the mitochondrial intermembrane space[1].
3-(N-Maleimidopropionyl)biocytin (0.5 mM; 30 min at 4 °C) does not label rat liver Tim44 in either intact or solubilized mitochondria, demonstrating that all cysteine residues of rat liver Tim44 are inaccessible to the reagent[1].
