In Vitro

3'-ONH2-dATP (0.25 mM; 20 min) led to wild-type ZaTdT exhibiting negligible catalytic activity for 3'-ONH2-dATP, with no detectable incorporation into CT-terminated synthetic DNA initiator strands under tested conditions[1].
3'-ONH2-dATP (0.25 mM; 20 min) led to engineered ZaTdT-R335A exhibiting 20-fold higher catalytic activity for 3'-ONH2-dATP than wild-type ZaTdT when tested with synthetic DNA initiator strands[1].
3'-ONH2-dATP (0.25 mM; 20 min) led to engineered ZaTdTR335L exhibiting a 2.6-fold higher catalytic activity for 3'-ONH2-dATP than ZaTdTR335A when tested with synthetic DNA initiator strands[1].
3'-ONH2-dATP (10 min per cycle) led to engineered ZaTdTR335L-K337G enabling efficient de novo enzymatic DNA synthesis with 3'-ONH2-dATP, supporting an average stepwise yield of 98.7% across 10 cycles and an 87.7% yield of full-length target synthetic DNA[1].
