7-Benzyloxy-4-(trifluoromethyl) coumarin (7-BFC) is a coumarin-based fluorescent substrate. 7-Benzyloxy-4-(trifluoromethyl) coumarin serves as a substrate for cDNA-expressed CYP1A2 and CYP3A4, and is metabolized into 7-hydroxy-4-trifluoromethylcoumarin (HFC). 7-Benzyloxy-4-(trifluoromethyl) coumarin supports metabolic studies of cytochrome P450 subtypes via its fluorescent metabolite.

7-Benzyloxy-4-(trifluoromethyl)coumarin (2-100 μM; 15 min) is metabolized to HFC in pooled liver microsomes with a mean Km of 8.3 μM and mean Vmax of 454 pmol/min/mg protein[1].
7-Benzyloxy-4-(trifluoromethyl)coumarin (20 μM; 15 min) metabolism in pooled liver microsomes is inhibited by CYP1A2 and CYP3A4 inhibitors and anti-CYP3A4 antibody, confirming primary catalysis by CYP1A2 and CYP3A4[1].
