AlkB (D135S/L118V), Nuclease-free (RNA m22G Demethylase (D135S/L118V)) is a nuclease-free modified demethylase. AlkB (D135S/L118V), Nuclease-free exhibits catalytic activity toward N2,N2-dimethylguanosine (m22G), N1-methylguanosine (m1G) and N1-methyladenosine (m1A). AlkB (D135S/L118V), Nuclease-free efficiently removes m1G and m1A modifications on RNA, and can also selectively convert m22G to N2-methylguanosine (m2G), thereby significantly reducing the level of m22G in transfer RNA (tRNA). AlkB (D135S/L118V), Nuclease-free greatly reduces sequencing bias and improves the efficiency of high-throughput tRNA sequencing.

Nuclease-free AlkB (D135S/L118V) demethylates the m22G substrate, which is a poor substrate for wild-type AlkB[1].
When incubated as a component of a three-enzyme mixture, AlkB (D135S/L118V), Nuclease-free (50-250 pmol; 30 min at 25°C), exhibits in vitro demethylase activity toward m1A- and m1G-modified RNA oligonucleotides[2].
When mixed with 500 pmol each of AlkB and AlkB D135S, AlkB (D135S/L118V), Nuclease-free (500 pmol; 30 min at 25°C), effectively removes m1A, m1G, m3C and m22G modifications from isolated zebrafish tRNA, and improves reverse transcription coverage in tRAM-seq analysis[2].
Nuclease-free AlkB (D135S/L118V) efficiently and selectively removes one methyl group from m22G in synthetic 9-mer RNA oligonucleotides to generate m2G, and shows no activity against m2G[3].
AlkB (D135S/L118V), Nuclease-free exhibits only limited demethylation activity towards m1G in synthetic 9mer RNA oligonucleotides, showing substrate selectivity distinctly different from that of the AlkB D135S mutant[3].
Nuclease-free AlkB (D135S/L118V) enhances the demethylation of m22G26 in tRNAs of HEK293T cells, reduces modification index values and improves tRNA sequencing efficiency, with the most significant effect observed on mitochondrial tRNAᴵˡᵉ[3].
