1. GPCR/G Protein Stem Cell/Wnt MAPK/ERK Pathway
  2. RXFP Receptor ERK
  3. B7-33

B7-33 is a single-chain relaxin mimetic and a selective relaxin receptor 1 (RXFP1) agonist. B7-33 phosphorylates ERK1/2 without inducing activation of the cAMP signaling pathway. B7-33 exhibits anti-fibrotic and cardioprotective activities. B7-33 can be used in the research of vascular diseases such as cardiomyopathy, myocardial infarction and fibrosis.

Description

B7-33 is a single-chain relaxin mimetic and a selective relaxin receptor 1 (RXFP1) agonist. B7-33 phosphorylates ERK1/2 without inducing activation of the cAMP signaling pathway. B7-33 exhibits anti-fibrotic and cardioprotective activities. B7-33 can be used in the research of vascular diseases such as cardiomyopathy, myocardial infarction and fibrosis[1][2].

In Vitro

B7-33 (10-100 nM) increases the survival rate of primary adult mouse cardiomyocytes subjected to simulated ischemia-reperfusion treatment via an ERK 1/2-dependent mechanism[1].
B7-33 (10-100 nM; 12 h) increases the survival rate of primary adult mouse cardiac fibroblasts subjected to simulated ischemia-reperfusion[1].
B7-33 (10-100 nM) dose-dependently increases the phosphorylation level of ERK1/2 in primary cardiomyocytes of adult mice[1].
B7-33 (100 nM; 24 h) reduces the expression of inflammasome marker ASC and endoplasmic reticulum stress marker GRP78 in adult primary mouse cardiomyocytes subjected to simulated ischemia-reperfusion[1].

In Vivo

B7-33 (0.25 mg/kg; i.p.; twice daily; 7 days) exerts a protective effect in a mouse model of ischemia-reperfusion[1].
B7-33 (0.25 mg/kg; s.c.; 7 days) exerts cardioprotective effects in a mouse model of cardiomyopathy[2].

Animal Model: CD1 (male, 6-8 weeks old; with ischemia-reperfusion model)[1]
Dosage: 0.25 mg/kg
Administration: i.p.; twice a day; 7 days
Result: Significantly reduced infarct size and preserved fractional shortening compared with vehicle.
Animal Model: 129sv mice (male, 14-16 weeks old, 25-30 g, Isoprenaline (HY-108353)-induced cardiomyopathy)[2]
Dosage: 0.25 mg/kg
Administration: s.c.; 7 days (days 7-14 post-injury)
Result: Reduced left ventricular (LV) fibrosis, normalized ISO-induced LV inflammation and cardiomyocyte hypertrophy, restored blood vessel density and aortic contractility in the model mice.
Restored α-SMA-stained smooth muscle blood vessel density, VEGF-stained endothelial blood vessel density and CD31-stained capillary density to the normal level.
Blunted the increased aortic contractility to phenylephrine caused by ISO injury.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
