BODIPY 558/568 C12 is a BODIPY dye. BODIPY dye is a small molecule dye with strong ultraviolet absorption ability, its fluorescence peak is relatively sharp, and the quantum yield is high. They are relatively insensitive to the polarity and pH of the environment and are relatively stable under different physiological conditions. Due to its structural asymmetry, BODIPY derives a variety of structural products. BODIPY lipid droplet dyes can well pass through the cell membrane into the cell, and localize the neutral lipids in the cell to specifically stain the lipid droplets, which can be used for labeling of live cells and fixed cells. Maximum excitation/emission wavelength: 558/568 nm.

Guide (The following is our recommended experimental protocol. This protocol serves as a reference guide only. Specific operations should be adjusted according to your actual needs.)
1. Preparation of Hoechst solution
1. Preparation of BODIPY558/568C12 solution
1.1 Prepare the storage solution
Prepare a 10 mM solution.
1.2 Preparation of the working solution
Dilute the storage solution with preheated serum-free cell culture medium or PBS to prepare a 1-10 μM BODIPY558/568C12 working solution.
Note: Please adjust the concentration of BODIPY558/568C12 working solution according to the actual situation, and prepare it as needed.
2. Cell staining (suspended cells)
2.1 Centrifuge to collect cells, add PBS for two washes, each for 5 minutes. Cell density is 1×106/mL
2.2 Add 1 mL of BODIPY558/568C12 working solution, incubate at room temperature for 5-30 minutes.
2.3 Centrifuge at 400 g for 3-4 minutes, discard the supernatant.
2.4 Add PBS for two washes, each for 5 minutes.
2.5 Resuspend the cells in 1 mL of serum-free medium or PBS, then observe using a fluorescence microscope or flow cytometer.
3. Cell staining (adherent cells)
3.1 Place adherent cells on a sterile cover glass.
3.2 Remove the cover glass from the culture medium, remove excess medium.
3.3 Add 100 μL of the dye working solution, gently shake to fully cover the cells, incubate for 5-30 minutes.
3.4 Remove the dye working solution, wash twice with culture medium, each for 5 minutes, and observe using a fluorescence microscope or flow cytometer.
Notes
1. Please adjust the concentration of BODIPY558/568C12 working solution and incubation time according to the actual situation.
2. It is recommended to conduct a positive control experiment. Place the control group cells in 30 μM oleic acid for 8 hours before proceeding with the subsequent experiments.
