Biological Activity

BODIPY 630/650X has advantages in live cell imaging, and its fluorescence signal can be clearly distinguished from cellular autofluorescence background[1].
Live cell imaging protocol[2][3]:
1. Preparation of stock solution
Dissolve BODIPY 630/650-X in DMSO to prepare a stock solution at a concentration of 1 mg/mL, and store at 4°C protected from light.
2. Preparation of working solution
Dilute the BODIPY 630/650-X stock solution with PBS to obtain a final working concentration of 5 μg/mL. The working solution should be freshly prepared and protected from light until use.
3. Cell staining
3.1 Wash cells twice with pre-warmed PBS to remove serum components.
3.2 Incubate cells with 5 μg/mL BODIPY 630/650-X working solution at room temperature for 25-30 min.
4. Washing
Wash cells 3-4 times with complete medium containing 5-10% serum or 1% BSA (or PBS) to quench unbound dye. Finally, wash once with live cell imaging buffer.
5. Imaging
Immediately acquire images using a confocal microscope or high-content imaging system. Excite with a HeNe laser (wavelength 633 nm) and collect emission using a ≥ 650 nm long-pass filter.
Note: The optimal dye concentration and incubation time may vary depending on cell type and application.
