Biological Activity
Guide (The following is our recommended protocol. This protocol only provides guidance and should be modified according to your specific needs).
1. Preparation of BODIPY 665/676 working solution
1.1 Preparation of stock solution
Dissolve 1 mg BODIPY 665/676 in 223 μL anhydrous DMSO to prepare a 10 mM stock solution.
Note: It is recommended to aliquot the BODIPY 665/676 stock solution and store at -20°C or -80°C, protected from light.
1.2 Preparation of working solution
Dilute the stock solution with pre-warmed serum-free cell culture medium or PBS to prepare a 1-10 μM BODIPY 665/676 working solution.
Note: Please adjust the concentration of the BODIPY 665/676 working solution according to actual conditions and prepare fresh before use.
2. UV-induced free radical reaction
2.1 Medium-chain triglyceride (MCT) oil and linseed oil (LSO).
a. Use a spectrofluorometer to measure fluorescence in the UV-induced free radical reaction.
b. Irradiate control oil (oil containing 1 μM BODIPY 665/676) and sample oil (oil containing 1 μM BODIPY 665/676 and 5.7 μM di-tert-butyl peroxide (DTBP)) for a fixed irradiation time.
c. Compared with the non-irradiated control oil, the fluorescence intensity measured at 675 nm in the sample decreases relatively, while the fluorescence intensity measured at 580 nm increases relatively, which is used to quantify the degree of oxidation. Measurements are repeated three times.

1. Please adjust the concentration of the BODIPY 665/676 working solution according to actual conditions.
2. Samples containing unsaturated fatty acids should be handled under nitrogen protection.
