DOPE-PEG2000-F3

DOPE-PEG2000-F3: surface functionalization of liposomes and nanoparticles for tumor-targeted drug delivery and tumor imaging, particularly for cancers with high nucleolin expression.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer that creates a steric shield reducing opsonization and RES clearance, prolonging circulation time;
(3) F3 peptide; a tumor-homing peptide derived from HMGN2 that specifically binds to nucleolin, a protein overexpressed on the surface of tumor cells and tumor vasculature endothelial cells (but not on normal cell surfaces), enabling active targeting of tumor tissues; F3 is also internalized by target cells via nucleolin-mediated endocytosis, facilitating intracellular drug delivery.
