CalFluor 647 azide is a fluorescent azide probe that can be activated via copper-catalyzed or metal-free click reactions. CalFluor 647 azide exhibits no fluorescence prior to reacting with alkynes. CalFluor 647 azide can be used for Cu2+ detection. The excitation wavelength of CalFluor 647 azide is 647 nm, with an emission range of 650 to 800 nm.

1. Working Solution Preparation
1.1 Working concentration: 2 μM.
1.2 Precautions: Adjust the working solution concentration as needed; prepare fresh before use.
2. Staining Procedure
2.1 Sample type description: Solution samples (including CuCl2 solution, artificial urine samples, bacterial suspension samples, and processed urine samples).
2.2 Incubation conditions: Use 2 μM CalFluor 647 azide working solution, incubate in the dark for 10 min with gentle shaking.
3. Detection and Analysis
3.1 Instrument type: Microplate reader.
3.2 Excitation/emission wavelengths: Excitation wavelength: 647 nm; emission wavelength: 650-800 nm.
3.3 Result analysis:
3.3.1 The higher the concentration of Cu2+ in the system, the stronger the fluorescence intensity.
3.3.2 In systems containing Cu2+, the higher the bacterial concentration, the weaker the fluorescence intensity.
3.3.3 After successful activation via click reaction, the dye emits red fluorescence.
