CDYL-IN-1 is a selective small-molecule inhibitor of CDYL with a KD value of 0.5 μM. CDYL-IN-1 improves renal dysfunction, pathological damage and renal tubular pyroptosis, inhibits FABP4-mediated ROS production, and alleviates renal tubular epithelial cell injury. CDYL-IN-1 is applicable to the research of acute kidney injury.

CDYL-IN-1 (D03) is non-toxic to TCMK-1 mouse renal tubular epithelial cells[1].
CDYL-IN-1 (D03) (100 μM; 12 h) alleviates injury and pyroptosis in cisplatin (HY-17394)-stimulated TCMK-1 mouse renal tubular epithelial cells by reducing CDYL expression[1].
CDYL-IN-1 (D03) (100 μM; 12 h) suppresses FABP4 expression in cisplatin-stimulated TCMK-1 mouse renal tubular epithelial cells[1].

Result:Reduced cisplatin-induced increases in kidney CDYL mRNA and protein levels.
Alleviated renal tissue damage observed via H&E staining.
Reduced serum creatinine levels.
Lowered mRNA levels of kidney injury markers NGAL and KIM-1.
Suppressed pyroptosis as shown by reduced mRNA levels of NLRP3 and GSDMD, and reduced protein levels of NLRP3, GSDMD-N, cleaved caspase 1, and cleaved IL-1β.
Detected no observable side effects on liver or kidney function, or in heart, liver, lung, or spleen tissues.
