Alkyne-PEG2000-CTP
Alkyne-PEG2000-CTP is a functionalized PEGylated lipid conjugate composed of three modular components: alkyne (propargyl/alkyne linker) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 2000 Da, and CTP (cardiotropic targeting peptide) (HY-P4094) as the terminal targeting/functional ligand. Alkyne-PEG2000-CTP does not contain a lipid anchoring group but instead provides a terminal reactive site for post-assembly ligand or probe conjugation on PEGylated nanoparticles.

Alkyne-PEG2000-CTP is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against brain endothelial cell receptors/CNS-specific transport mechanisms.
Composition and role of each component:
1. PEG2000 (polyethylene glycol, MW 2000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
2. CTP: Targeting ligand-can be reversibly linked (e.g., via enolase, thiol groups) to a carrier (e.g., miRNA), thereby enabling specific delivery to cardiomyocytes rather than all other organs.
