In Vivo

IR-19-Py LNP (1 mg firefly luciferase mRNA; administered via nebulization; once every 72 h (repeated administration); single administration) delivers nebulized mRNA to the lungs and nasal cavities of mice, enabling long-term expression in the nasal cavity^[1].

Animal Model:

C57BL/6J (female, 6-8 weeks old)^[1]
Dosage:

1 mg firefly luciferase mRNA (single dose; 3 total doses)
Administration:

nebulized; every 72 hours (repeat dosing); single dose (initial)
Result:

Lasted at least 48 hours post-nebulization in lung protein expression.
Exhibited prolonged nasal luciferase expression.
Showed no loss in lung luminescence over three repeat doses administered every 72 hours.
Showed a dose-dependent increase in lung luminescence with increasing nebulized mRNA dose.
