Biological Activity
CMV pp65 (123-131) HLA-B*35:01/IPSINVHHY-specific CD8+ T cells (CFSE 5 μM; IL-2 10 U/mL; stimulated with allogeneic PBMCs; 8 days) exhibit proliferative responses to HLA-B51+ and/or HLA-B58+ allogeneic cells in 23% (3/13) of tetramer-positive healthy donors, with HLA-B51-induced proliferation being stronger than that induced by HLA-B58[1].
CMV pp65 (123-131) HLA-B*35:01/IPSINVHHY-specific CD8+ T cell clones (E:T ratios of 30:1, 10:1, 1:1, and 0.1:1; IL-2 10 U/mL; 4 h) lyse HLA-B51+ PHA blasts, EBV-LCLs, PTECs, and HUVECs, whereas HLA-B58/B57 cross-reactive clones cannot effectively lyse the corresponding tested target cells[1].
CMV pp65 (123-131)/HLA-B*35:01 (IPSINVHHY)-specific CD8+ memory T cell clones exhibit shared allo-HLA cross-reactivity with highly conserved usage of TCR Vα and Vβ. Only minor CDR3β amino acid differences exist among distinct clones, and these differences can affect the cross-recognition patterns of HLA-B51 versus HLA-B58/B57[1].
CMV pp65 (123-131) HLA-B*35:01/IPSINVHHY-specific CD8+ T cell clones (5 × 103 T cells; 5 × 104 EBV-LCLs; IL-2 10 U/mL; 24 h) produce IFN-γ in response to HLA-B51+, HLA-B58+ and HLA-B57+ EBV-LCLs, with stronger responses induced by HLA-B51 and HLA-B58 than by HLA-B57[1].

HLA-B58/B57-cross-reactive clones did not efficiently lyse the tested HLA-B58/B57+ target cells; HLA-B58+ HUVECs were not tested.
