Description

Cy5-ZW2-617 is a PSMA ligand with a IC50 of 4.39 nM. Cy5-ZW2-617 specifically binds to PSMA and is thereby internalized by PSMA-positive cells. Cy5-ZW2-617 shows uptake in PSMA-positive tumor cells and tumor xenografts in mice. Cy5-ZW2-617 can be used for the research of prostate cancer[1].

In Vitro

Cy5-ZW2-617 (Compound 10) (2.5-10 μM; 2 h) is efficiently and specifically endocytosed by PSMA-positive PC3-PIP and LNCaP cells, and then mainly localizes to the perinuclear compartment, while no uptake is observed in PSMA-negative PC3 flu cells[1].

Immunofluorescence[1]

Cell Line: PSMA-positive PC3-PIP, LNCaP cells
Concentration: 2.5 μM or 10 μM
Incubation Time: 2 h
Result: Showed efficient internalization into PSMA-positive PC3-PIP and LNCaP cells, with highest fluorescence intensity observed in perinuclear compartments.
In Vivo

Cy5-ZW2-617 (20 nmol; retro-orbital injection; single dose) is specifically taken up in PSMA-positive prostate cancer xenograft mice[1].

Animal Model: athymic nude-Fox1nu (male, 6 weeks old, subcutaneous xenograft modeling with PSMA-positive PC3-PIP and PSMA-negative PC3 flu cells)[1]

Result: Exhibited high, specific uptake in PSMA-positive PC3-PIP tumors, with low uptake in PSMA-negative PC3 flu tumors.
